Identificación de los principales alcaloides de Cinchona micrantha por UHPLC-MS/MS

Abstract
El objetivo de esta investigación fue identificar los principales alcaloides de Cinchona micrantha mediante cromatografía líquida usando el equipo de UHPLC-MS/MS, en el procesamiento de los datos se usó el software Masslynx (Water Corporation). La muestra de estudios fue el extracto acuoso acidulado de Cinchona micrantha. La elución del gradiente se efectuó usando la fase móvil A: buffer fosfato de potasio 0.06M pH=3 y B: acetonitrilo (95:5 v/v), la fase estacionaria fue la columna C18 ACQUITY UPLC® BEH C18 1.7μm 2.1 x 100 mm. Se observaron los espectros de masa en modo ion positivo, identificándose según el análisis respectivo los espectros de los siguientes alcaloides: cinconidina con tR de 17.66 min, el ion precursor de 295.99 m/z y los fragmentos 295, 277, 168, 130, 81, 79; cinconina con un tR de 17.76 min, el ion precursor de 296,19 m/z y los fragmentos 296, 278, 168,156,142; dihidrocinconidina con un tR de 18.22 min, el ion precursor de 297 m/z y los fragmentos 297, 279, 83, 81; quinina con un tR de 18.32 min, el ion precursor de 325.48 m/z y los fragmentos 307, 265, 251, 210, 198, 172, 160, 108, 81, 79; quinidina con un tR de 18.40 min y el ion precursor de 325.18 m/z; dihidroquinina con un tR de 18.77 min, el ion precursor de 327.78 m/z y los fragmentos 309, 174, 162, 110, 83, 81. Se logró identificar seis principales alcaloides de Cinchona micrantha por UHPLC-MS/MS.
ABSTRACT The objective of this research was to identify the main alkaloids of Cinchona micrantha by liquid chromatography using the UHPLC-MS/MS equipment, in the data processing the Masslynx software (Water Corporation) was used. The study sample was the acidified aqueous extract of Cinchona micrantha. The elution of the gradient was carried out using the mobile phase A: 0.06M potassium phosphate buffer pH=3 and B: acetonitrile (95:5 v/v), the stationary phase was the ACQUITY UPLC® BEH C18 1.7μm 2.1 x C18 column. 100mm Mass spectra were observed in positive ion mode, identifying the spectra of the following alkaloids according to the respective analysis: cinchonidine with tR of 17.66 min, the precursor ion of 295.99 m/z and the fragments 295, 277, 168, 130, 81, 79 ; cinchonine with a tR of 17.76 min, the precursor ion of 296.19 m/z and fragments 296, 278, 168,156,142; dihydrocinchonidine with a tR of 18.22 min, the precursor ion of 297 m/z and fragments 297, 279, 83, 81; quinine with a tR of 18.32 min, the precursor ion of 325.48 m/z and fragments 307, 265, 251, 210, 198, 172, 160, 108, 81, 79; quinidine with a tR of 18.40 min and the precursor ion of 325.18 m/z; dihydroquinine with a tR of 18.77 min, the precursor ion of 327.78 m/z and fragments 309, 174, 162, 110, 83, 81.
