Efecto in vitro del extracto metanólico de la cáscara de Punica granatum sobre la supervivencia de Streptococcus agalactiae

dc.description.abstractEl presente trabajo tuvo como objetivo evaluar la supervivencia de Streptococcus agalactiae ATCC 13813 frente a tres diferentes concentraciones de extracto metanólico de la cáscara de Punica granatum. Para la obtención del extracto se secó las cáscaras y se molió hasta obtener un polvo fino, el cual se maceró en metanol al 80% para luego llevar a rotavapor y posteriormente a la estufa para secado, y para determinar la sensibilidad (supervivencia) antibacteriana, se emplearon cultivos de Streptococcus agalactiae que fueron reactivados en Caldo TSA por 24 horas a 37°C y estandarizados al tubo N° 1 del Nefelómetro de Mac Farland (3x108UFC/mL). La supervivencia bacteriana frente al extracto se determinó utilizando el método de dilución en tubo en caldo Mueller Hilton, a 37°C y a concentraciones de 0.10, 0.25 y 1.20 mg/mL; asimismo un control conteniendo caldo Mueller Hinton con inóculo bacteriano. Se sembraron por superficie en placas Petri con Agar Mueller Hinton, La supervivencia se midió mediante el recuento en placas comparadas con un control, esto se realizó por triplicado. Concluyéndose que el extracto metanólico de Punica granatum mostró una actividad antibacteriana sobre Streptococcus agalactiae en la concentración de 1.20 mg/mL sobre las concentraciones de 0.10 mg/mL, 0.25 mg/mL y el control.
dc.description.abstractThe objective of this work was to evaluate the survival of Streptococcus agalactiae ATCC 13813 against three different concentrations of methanolic extract from the peel of Punica granatum. To obtain the extract, the shells were dried and ground until a fine powder was obtained, which was macerated in 80% methanol and then taken to a rotary evaporator and later to the oven for drying, and to determine the antibacterial sensitivity (survival). Streptococcus agalactiae cultures were used, which were reactivated in TSA Broth for 24 hours at 37°C and standardized to tube No. 1 of the Mac Farland Nephelometer (3x108UFC/mL). Bacterial survival against the extract was determined using the tube dilution method in Mueller Hilton broth, at 37°C and at concentrations of 0.10, 0.25 and 1.20 mg/mL; also a control containing Mueller Hinton broth with bacterial inoculum. They were surface plated in Petri dishes with Mueller Hinton Agar. Survival was measured by plate counts compared to a control, this was done in triplicate. It was concluded that the methanolic extract of Punica granatum showed antibacterial activity against Streptococcus agalactiae in the concentration of 1.20 mg/mL over the concentrations of 0.10 mg/mL, 0.25 mg/mL and the control.
