Determinación de la actividad antimicrobiana in vitro de las trampas extracelulares de neutrófilos purificados de pacientes con tuberculosis

Abstract
Objetivo: Determinar la actividad antimicrobiana in vitro de las trampas extracelulares de neutrófilos purificados de pacientes con tuberculosis. Método: Este trabajo se fundamenta en el uso del ensayo de susceptibilidad a fármacos en caldo de observación microscópica (MODS), el cual permite la detección Mycobaterium tuberculosis y permite evaluar la susceptibilidad frente a los antimicrobianos. Las Trampas extracelulares de neutrófilos (NETs) serán obtenidos por purificación de muestras de esputo de pacientes con tuberculosis, por centrifugación. Se trabajará cuatro diluciones de trampas extracelulares de neutrófilos purificado (25%, 50%, 75%, 90%), La evaluación de eficacia del extracto de NETs purificado frente a Mycobacterium tuberculosis se realizó en una placa de poliestireno estéril de 24 pocillos de fondo plano la cual contiene las diferentes diluciones de NETs a las que posteriormente se inoculó la bacteria. Luego se contrastó las observaciones obtenidas de los pozos control vs los pozos que contenían las diferentes concentraciones de NETs.
Resultados: No se observó efecto antimicrobiano en ninguna de las concentraciones. Es decir, todas las concentraciones (25, 50, 75, 90%) arrojaron resultados positivos, no hallándose diferencias significativas con el grupo control (pozos de Mycobacterium tuberculosis sin NETs).
Conclusión: Las trampas extracelulares de neutrófilos (NETs) purificados no tienen eficacia antimicrobiana frente a cepas de Mycobacterium tuberculosis.

Method: This work is based on the use of the MODS methodology, which allows the detection of M. tuberculosis and allows the evaluation of susceptibility to antimicrobials. The NETs will be obtained by purification of sputum samples from patients with tuberculosis, by centrifugation. Four dilutions of purified NETs will be used (25%, 50%, 75%, 90%). The efficacy evaluation of the purified NETs extract against Mycobacterium tuberculosis was carried out in a 24-well flat bottom sterile polystyrene plate which contains the different dilutions of NETs at which the bacteria were subsequently inoculated. Then, the observations obtained from the control wells were contrasted against the wells that contained the different concentrations of NETs. Results: No antimicrobial effect was observed in any of the concentrations. That is, all concentrations (25, 50, 75, 90%) yielded positive results, not finding significant differences with the control group (Mycobacterium tuberculosis wells without NETs). Conclusion: Purified neutrophil extracellular traps (NETs) have no antimicrobial efficacy against Mycobacterium tuberculosis strains.
