Efecto de medios de cultivo sobre la producción de enzimas y delta-endotoxina por Bacillus thuringiensis

Abstract
El objetivo del presente trabajo fue determinar el efecto del Caldo Nutritivo y Caldo Nutritivo suplementado con Quitina Coloidal al 2%, sobre la producción de proteasas, quitinasas y delta-endotoxinas por Bacillus thuringiensis con código Bt-14-NP proporcionado por empresa Camposol, se preparó el inóculo bacteriano a la concentración de 3x108 células/mL, luego se procedió a inocular 1mL a botellas de vidrio con 99mL del medio de cultivo previamente esterilizado; se utilizó como control Caldo Quitina Coloidal. Las evaluaciones de producción de proteasas, quitinasas y delta-endotoxinas, así como las UFC/mL y número de esporas, se realizaron a las 12, 24, 48, 60, 72, 84, 96, 108, 120, 132 y 144 horas de cultivo; la incubación se realizó a 29.5°C y 200 rpm de agitación. Los resultados evidencian que el Caldo Nutritivo tiene efecto positivo sobre la producción de proteasas entre 60 y 84 horas y de delta-endotoxina a las 72 h de incubación; el Caldo Nutritivo suplementado con 2% de Quitina Coloidal favorece la producción de quitinasas a las 72 horas de incubación. Así mismo, el mayor crecimiento y la esporulación se ve favorecida en Caldo Nutritivo.
The objective of this work was to determine the effect of Nutrient Broth and Nutrient Broth supplemented with 2% Colloidal Chitin, on the production of proteases, chitinases and delta-endotoxins by Bacillus thuringiensis with code Bt-14-NP provided by Camposol company. The bacterial inoculum was prepared at a concentration of 3x108 cells/mL, then 1 mL was inoculated into 250 mL capacity glass bottles with 99 mL of the previously sterilized culture medium; Colloidal Chitin Broth was used as a control. The evaluations of protease, chitinase and delta-endotoxin production, as well as CFU/mL and number of spores, were carried out at 12, 24, 48, 60, 72, 84, 96, 108, 120, 132 and 144 hours of crop; Incubation was carried out at 29.5°C and 200 rpm shaking. The results show that the Nutrient Broth has a positive effect on the production of proteases between 60 and 84 hours and delta-endotoxin at 72 hours of incubation; Nutrient Broth supplemented with 2% Colloidal Chitin favors the production of chitinases after 72 hours of incubation. Likewise, greater growth and sporulation is favored in Nutritive Broth.
