dc.descriptionIn the present research work, cytotoxic effect of cadmium and lead in apical cells of the secondary roots of Vicia faba "bean" seedlings was determined. For this, we worked with 27 seedlings of Vicia faba "haba" with 10 days of vegetative development and individually each covered by cotton and placed in disposable cups, embedded with distilled water free of lead and cadmium metals. Four solutions of cadmium and four of lead plus a control solution were prepared, with three repetitions for each treatment; The imbibition of each metal was performed for 3 days. The apexes were cut by immersing in 1N hydrochloric acid, then stained with 2% acetic Orcein and observed under a microscope; the mitotic index was calculated and cellular (chromosomal) abnormalities were observed. An analysis of variance (ANOVA) and multiple range test was performed using the method of significant minimum difference HSD TUKEY. The results showed that there is a statistically significant difference between treatments (P> 0.05). The concentration of cadmium where there was a higher MI was 1 ppm, while for lead it was 5ppm. Observing cell abnormalities: in anaphase, showing bridges between chromosomes, nuclear lesions and chromosome breakage in anaphase and metaphase. The results showed that cadmium was the metal that caused a greater cytotoxic effect in the cells of the mitotic division of Vicia faba "haba", this is a species that offers an experimental model to evaluate the cytotoxic effect, in this case of cadmium and lead, which generated cellular (chromosomal) abnormalities at all concentrations.es_PE
dc.description.abstractEn el presente trabajo de investigación se determinó efecto citotóxico de cadmio y plomo en células apicales de las raíces secundarias de plántulas de Vicia faba "haba". Para ello se trabajó con 27 plántulas de Vicia faba "haba" con 10 días de desarrollo vegetativo y de forma individual cada una cubierta por algodón y colocadas en vasitos descartables, embebidas con agua destilada libre de metales plomo y cadmio. Se prepararon 4 soluciones de cadmio y cuatro de plomo más una solución control, con tres repeticiones para cada tratamiento; se realizó la imbibición de cada metal durante 3 días. Se cortaron los ápices sumergiéndose en ácido clorhídrico 1N, posteriormente fueron teñidos con Orceina acética al 2% y se observaron al microscopio; se calculó el índice mitótico y se observaron anomalías celulares (cromosómicas). Se realizó un análisis de varianza (ANOVA) y prueba de rangos múltiples mediante el método de diferencia mínima significativa HSD TUKEY. Los resultados mostraron que si existe una diferencia estadísticamente significativa entre los tratamientos (P>0,05). La concentración de cadmio donde se presentó un mayor IM fue de 1 ppm, mientras que para plomo fue 5ppm. Observándose las anomalías celulares: en anafase, evidenciando puentes entre cromosomas, lesiones nucleares y rompimiento de cromosomas en anafase y metafase. Los resultados evidenciaron que cadmio fue el metal que ocasiono un mayor efecto citotoxico en las células de la división mitótica de Vicia faba "haba", esta es una especie que ofrece un modelo experimental para evaluar el efecto citotóxico, en este caso del cadmio y plomo, el cual generó anomalías celulares (cromosómicas) en todas las concentraciones.es_PE
