Caracterización fisicoquímica de enalapril maleato 10 mg y losartán potásico 50 mg en tabletas adquiridas por el estado peruano

dc.descriptionABSTRACT The objective of the research was to determine the physicochemical characteristics of enalapril maleate 10 mg and losartan potassium 50 mg tablets acquired by the Peruvian state. Tablets of reference drug (Lotrial® and Cozaar®), tablets acquired by the Peruvian state and of private sale were used. In addition, we worked with secondary standard Sigma-Aldrich® and active pharmaceutically ingredient of the national industry. The dissolution profiles at pH 1.2, 4.5 and 6.8 were evaluated, between 3 and 30 minutes, using the USP method II. The identification of functional groups was performed by infrared spectroscopy (ATR-FTIR) in the range of 4000 to 450 cm-1 and the melting points by differential scanning calorimetry (DSC) in the range of 30 to 300 °C with ramp of 10 °C/min. The best fit kinetics for the tablets of enalapril maleate 10 mg was Cubic root model and the Kd ranged between 0.0478 and 0.1959 mg1/3/min and for losartan potassium 50 mg tablets, Order one model, with values of Kd between 0.0021 and 0.1387 min-1. In all cases, the dissolution profiles of the test tablets were different from the reference. Vibration signals were obtained in IR of 3211 cm-1 (N-H), 1750 cm-1 (C=O, ester), 1725 cm-1 (C=O, acid) and 1645 (C=O, amide) for enalapril maleate; for losartan potassium at 3194-3182 cm-1 (O-H), 1579-1578 cm-1 (C=N) and 762-763 cm-1 (C-Cl). In the DSC, melting points of 151 °C (enalapril maleate) and 269-270 °C (losartan potassium) were obtained. It is concluded that the tablets of enalapril maleate 10 mg and losartan potassium 50 mg acquired by the Peruvian state have physicochemical characteristics in both the dissolution profile, ATR-FTIR and DSC differentiated to the reference drug.es_PE
dc.description.abstractRESUMEN La investigación tuvo por objetivo determinar las características fisicoquímicas de enalapril maleato 10 mg y losartán potásico 50 mg en tabletas adquiridas por el estado peruano. Se emplearon tabletas de medicamento referencia (Lotrial y Cozaar), tabletas adquiridas por el estado peruano y de venta privada. Además, se trabajó con estándar secundario Sigma-Aldrich e ingrediente farmacéuticamente activo de la industria nacional. Se evaluaron los perfiles de disolución a pH 1,2; 4,5 y 6,8, entre 3 y 30 minutos, utilizando el método II USP. La identificación de los grupos funcionales se realizaron mediante espectroscopia infrarroja (ATR-FTIR) en el rango de 4000 a 450 cm-1 y los puntos de fusión por calorimetría diferencial de barrido (DSC) en el rango de 30 a 300 °C con rampa de 10 °C/min. La cinética de mejor ajuste para las tabletas de enalapril maleato 10 mg fue modelo Raíz cúbica y los Kd fluctuaron entre 0,0478 y 0,1959 mg1/3/min y para las tabletas de losartán potásico 50 mg, modelo Orden uno, con valores de Kd entre 0,0021 y 0,1387 min-1. Para todos los casos los perfiles de disolución de las tabletas ensayo fueron diferentes al de referencia. Se obtuvieron señales vibracionales en IR de 3211 cm-1 (N-H), 1750 cm-1 (C=O, éster), 1725 cm-1 (C=O, ácido) y 1645 cm-1 (C=O, amida) para enalapril maleato; para losartán potásico a 3194-3182 cm-1 (O-H), 1579-1578 cm-1 (C=N) y 762-763 cm-1 (C-Cl). En el DSC se obtuvieron puntos de fusión de 151 °C (enalapril maleato) y 269-270 °C (losartán potásico). Se concluye que las tabletas de enalapril maleato 10 mg y losartán potásico 50 mg adquiridas por el estado peruano presentan características fisicoquímicas tanto en perfil de disolución, ATR-FTIR y DSC diferenciadas al medicamento referencia.es_PE
